Elongation Factor- Thermo Unstable (EF-Tu) in pSB1C3

EF-Tu (elongation Factor thermo unstable) is a prokaryotic protein that aids the binding of an aminoacyl-tRNA (aa-tRNA) to the ribosome. Thus EF-Tu aids the elongation of peptide chains during protein synthesis [1]. This part is codon optimized for use in Escherichia coli , contains an N-terminal hexahistidine tag for purification and has a T7 Promoter, medium, strength RBS and double terminator. This part was originally designed by the 2017 Lethbridge iGEM team (http://2017.igem.org/Team:Lethbridge) and was initially characterized in the pUC57 plasmid (https://parts.igem.org/Part:BBa_K2443027).

This part was improved by placing the part in the pSB1C3 plasmid.

Figure 1- An overexpression gel of EF-Tu run on a 12% SDS-PAGE. Samples were taken at point of induction and in 1 hour increments thereafter. Although induction is weak, the protein is seen at the 45kDa range.
